研究略歴
Research Profile
大学院時代、私は天然物有機化学を専門としていました。橘和夫教授および村田道雄助教授(東京大学)の下で海洋微生物(渦鞭毛藻)から得られる天然毒の構造解析を行い、NMRとの格闘の日々を送っていました。当時開発した方法はJ-based configuration analysis (JBCA法)と名付けられ(図1)、現在では鎖状天然有機化合物の立体化学決定法のスタンダートとなっています。
博士取得後は、東大農学部の堀之内末治教授ならびに吉田稔助教授(現理研主任研究員)の下でポスドクを行い、分子生物学やケミカルバイオロジーに関する研究の機会を得ました。その後、平成11年に大阪大学で助手に採用され、村田道雄教授の下で准教授まで務めました。途中1年間MITのGriffin研で固体NMRを学ぶ機会を得ました。
大阪大学時代には、膜に作用する天然分子アンフォテリシンBの研究から始まり、バイセルや脂質ラフト、さらには膜タンパク質まで扱うようになりました。それに伴い、分析手法も、溶液および固体NMRから表面プラズモン共鳴、熱測定、蛍光顕微鏡、さらには共同研究でありますが、膜タンパク質の膜環境下での結晶化やX線解析なども行う機会に恵まれました(図2)。そのような研究が幸いし、2014年に九州大学で脂質膜を分析化学的に研究する生体分析化学研究室を主宰することになりました。現在は、「研究内容」で述べたように、生体膜の統合分析プラットフォームを構築し、生体膜を多角的かつ統合的に解析していくことを目標に研究を行っています。

I started my career as a natural product chemist, having studied marine natural toxins. My Ph.D. research, which was carried out under the supervision of Prof. K. Tachibana and Assoc. Prof. M. Murata, was on the development of J-Based Configuration Analysis (JBCA) method (Figure 1), which is now a standard method for determining relative configurations in acyclic natural producs. After two years of postdoctoral research in antibiotic biosynthesis and chemical biology with Prof. S. Horinouchi and Assoc. Prof. M. Yoshida at the University of Tokyo, I was appointed as an Assistant Professor (1999), and then an Associate Professor (2010) at Osaka University, where I worked with Prof. M. Murata. In 2000, I spent a year as a visiting scientist in the solid-state NMR laboratory of Prof. R. G. Griffin at MIT.
After moving to Osaka University, I first studied amphotericin B, which is a clinically important antifungal drug known to form ion-channels in fungal membranes, using solid state NMR and other analytical methods. Then I fell deep inside the membrane and was devoted to membrane studies including bicelles, lipid rafts, and membrane proteins (Figure 2).
In 2014, I was promoted to a Professor at Kyushu University and launched Laboratory of Bioanalytical Chemistry. My research interest is in the multifaceted analysis of membrane systems, including membrane proteins and lipid rafts, via constructing "integrated analytical platform".
